Yes, squads can be changed - however one limitation is that only one player who has played in European competition previously this season for another club can be added to the CL squad for the latter stages - which is why Real Madrid had a problem when they bought Lassana Diarra and Klass Jan Huntelaar a couple of years back - Lass had already played for Portsmouth and KJH for Ajax, and they couldn't register both into their CL squad.

I believe up to 25 players can be registered, but there's also a B-list for players under 21 years of age that can be submitted, and I don't know of any numerical limits to that list.
